Methods: PNU-282987 was administered to mice that received recombinant IL-33 or AA intranasal challenges. Lung histological analysis and flow cytometry were performed to determine airway inflammation and the infiltration and activation of ILC2s. The previously published a7nAChR agonist GTS-21 was employed as a comparable reagent. ILC2s were isolated from murine lung tissue and cultured in vitro in the presence of IL-33, IL-2, and IL-7 with/without either PNU-282987 or GTS-21. The expression of the transcription factors GATA3, IKK, and NF-kB were also determined. Results: PNU-282987 and GTS-21 significantly reduced goblet cell hyperplasia in the airway, eosinophil infiltration, and ILC2s numbers in BALF, following IL-33 or AA challenge. In vitro IL-33 stimulation of isolated lung ILC2s showed a reduction of GATA3 and Ki67 in response to PNU-282987 or GTS-21 treatments. There was a significant reduction in IKK and NF-kB phosphorylation in the PNU-282987–treated group when compared to the GTS-21–treated ILC2s. Conclusion: PNU-282987 inhibits ILC2-associated airway inflammation, where its effects were comparable to that of GTS-21.]]> Tue 15 Nov 2022 10:22:01 AEDT ]]> Inhibition of β-catenin/CBP signalling improves airway epithelial barrier function and suppresses CCL20 release https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:40080 Tue 05 Jul 2022 08:35:39 AEST ]]> Airway monocyte modulation relates to tumour necrosis factor dysregulation in neutrophilic asthma https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:45375 Thu 27 Oct 2022 16:54:22 AEDT ]]> Using fractional exhaled nitric oxide to guide asthma therapy: design and methodological issues for ASthma TReatment ALgorithm studies https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:8263 Sat 24 Mar 2018 08:40:37 AEDT ]]> Coronary-bronchial blood flow and airway dimensions in exercise-induced syndromes https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:1971 Sat 24 Mar 2018 08:33:15 AEDT ]]> The 'classical' ovalbumin challenge model of asthma in mice https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:5604 Sat 24 Mar 2018 07:49:22 AEDT ]]> The IL-3/IL-5/GM-CSF common β receptor plays a pivotal role in the regulation of Th2 immunity and allergic airway inflammation https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:5180 Sat 24 Mar 2018 07:47:49 AEDT ]]> Effect of obesity on airway and systemic inflammation in adults with asthma: a systematic review and meta-analysis https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:53538 Mon 04 Dec 2023 15:44:36 AEDT ]]> Supraglottic airway devices for administration of surfactant to newborn infants with respiratory distress syndrome: a narrative review https://ogma.newcastle.edu.au/vital/access/ /manager/Repository/uon:39809 Fri 24 Jun 2022 09:07:06 AEST ]]>
